In this scene, Pooh has already undertaken a long, exhausting, and dangerous search for his dear friend with no clear indicator where he is other than a false clue given by Owl that he is at a place called "Skull" (which later turned out to be a mispronunciation for the word, "school"). However, Pooh and his friends can't seem to find him despite their best efforts and late at night, he looks up to the Moon and stars and pleads with them to help him find Christopher Robin. This is probably the best known song from the movie as it beautifully captures the character in despair while also hinting at a glimmer of hope.
