I find the idea of Transgender Donatello interesting. It could play in to the fact that they do not like being hugged very long in the new show.

However, I would like to suggest an artist challenge to you- feel free to decline if you dislike the idea.

Breasts are for mammals and humans. As a turtle, Donatello would not have them no matter what gender they are.

So instead, how else could you show that Donatello was born female but identifies as male?
What female turtle qualities must Donatello overcome in order to be seen as a male?

Perhaps when the four turtles hit puberty, Raph, Leo, and Mikey's tails all grew long and tapered. But Donatello's stayed short and stubby.

Turtles have a stronger sense of smell than a dog!
Donatello would hate that he smells different than his bros, and that during turtle breeding season he would smell very female (and smells very tempting).

Also, while breeding season would makes his brothers cranky and restless, Donatello would feel hungry for strange foods (like eggshells for more calcium) bloated, and needy for attention (even hugs!).

A trans-male human is embarrassed by the fact that they have to deal with menstruation. A trans-male turtle would be embarrassed that they lay eggs.

Male turtles churr, but female turtles make a high pitched chirp. So maybe when frightened or excited, sometimes Donatello forgets to lower their voice and it sounds too high to be a male?

Donatello could also be upset that he can not gain as much muscle mass as his brothers, and society sees big muscles as masculine. But Donatello has slender, graceful yet curvy legs and wider (but not fat) hips that look feminine that he hates!

Splinter seems to call the turtles by the color of their masks.
Maybe Splinter used to dress Donatello in magenta or lavender- which are shades of reddish-purple that are for girls. And maybe Donatello started dying their masks the darker shade of bluish-purple that they are now, which is seen as masculine.
Perhaps Splinter is not understanding, and refuses to say they name "Donatello" because he knows his daughter is a "Donatella" so calling them "Purple" is a compromise. The new version of Splinter in "Rise of the TMNT" seems like a poor father who is insensitive, so it would be suitable to his character.

Perhaps you could even draw out scenes from the turtles' childhood; the past when "Donnie" was still called "Donna", and show how lonely and out of place they felt being the only sister in a family of brothers.
How "Donna" hated being gifted with used rag-dolls and chipped teapot sets to play with, while her brothers got cool toy cars with missing wheels or patched up sports balls.
Or how uncomfortable Donna felt being dressed up in a Japanese kimono bow around the waist that felt big and bulky and was no good to run and play in, but her brothers only had to wear normal belts that had more freedom of movement and did not come undone or become a tripping hazard if they played too rough.

Perhaps Splinter always thought that girls should like pictures of flowers and reading fairytales about princesses or learning how to bake cakes.
But Donna liked pictures of dinosaurs and reading about car repair and space travel and wanted to learn how to build a robot! (Mikey wanted to learn how to make cakes.)

Show a childhood where Splinter keeps making assumptions about what the purple turtle should like and how they should behave because they are a girl. And show how, as Donna gets older, grows to hate these gender stereotypes and hates being associated with them because they all feel wrong.
Build it up to the day Donna finally realizes that the reason these stereotypes all feel so awful and wrong is because Donna is actually a "Donnie", he just has the wrong body.

Have you thought about how other characters will react?

I think April will treat Donnie as a male.

I think Mikey would be understanding and call Donnie a boy but sometimes he says things like "You are so pretty- and by pretty I mean handsome!" Or "You are so cute- and that is ok because I am a guy and I am adorable"

The new Raph is more sensitive and open with his feelings than previous versions- I think he would want to be supportive and accepting, but he still makes mistakes sometimes and will accidentally use female pronouns or call Donnie "Donna" or "sis" instead of "bro" and he instantly feels awful and apologizes over and over again until Donnie says "Enough! It's fibe, just change the subject already!"

This new Leo is more sarcastic and has an ego- I think most of the time he would not care, but if he was in a cranky mood or in an arguement with Donnie, Leo would mutter a bad joke about it being "thst time of the month" to explain Donnie's disagreeability, or say something snarky like "Stop acting like such a girl." But eventually he would apologize because even if he doesn't fully understand why Donnie says he is a boy when he was born a girl, Leo still cares enough not to want to damage Donnie's feelings.

Sometimes families say hurtful things but it is important to apologize.

I think Splinter would not like it, he knows he has 3 sons and 1 daughter!
After puberty, they came to a bitter truce- Splinter calls the turtles by their colors so that the purple one will not yell about being called "Donatella" and to avoid a lecture about transgender issues. Splinter thinks it is stupid for his daughter to pretend to be a man, and Donnie is frustrated with his father's stubborness. But many years later, for Donnie's 18th birthday he gets a wonderful gift- Splinter finally calls him "Donatello, my son."

Your work will have more dynamic and depth if there are different reactions from the characters. People who don't want to understand, people who do, people who want to but makes mistakes, these are things that people can empathize with and will make your art and stories feel alive. It creates empathy.

The good thing about bad guys is that most people can not tell the difference between male and female turtles. All villains would just assume that all four turtles were born male, and Donatello is fine with that!

If you don't like these suggestions I will understand. I will not take offense. I hope I have not offended you. I look forward to seeing more of your art

Oh, sorry if i'm responding late. I already know some of those facts, but I'm very happy you bring those on! Yeah, I liked some of those ideas really much! I have read some of yours fanfics and I can say that you understands more the turtles than me, it's pretty cool because I really need help a little so you helped me and I really liked those suggestions. Thank you.
Donnie too would be a little frustrated because of the little "elevetion" he have on his torso, he knows his brothers have one too but sometimes he will think more of mammals than reptiles and that would bring some of his ego down, so he decide to look more "manly" and draw some eyebrows, just to look more safe with himself.
(My english is very bad, I'm sorry)

Oh! You could totally tie this in with existing episodes, like "Donnie's Gifts"!

Like, imagine when Donnie first started drawing eyebrows on their mask- they thought it would look more manly like Lou Jitsu!

But Leo wasn't thinking and said something like "Seriously? You are drawing on eyebrows? Isn't that what desperate girls on instagram do?" And there was a pause before Leo's eyes widened realizing what he said, and Raph is able to restrain Donnie from using the tech-bo to punish Leo, but Donnie swore revenge one day-

So Donnie creates a shock collar- one that does not just zap his brother for lame jokes- but gives Leo a nasty zap everytime he accidently uses the wrong gender pronoun or says Donna instead of Donnie.

Leo is not trans-phobic though, he just does not understand because he thought his sister was perfect just the way she is. But he does not hate Donnie for identifying as male. Leo just has a hard time understanding and remembering, so Donnie does not hate Leo either- just gets very annoyed with him!
